In vitro colonization and increase in nitrogen fixation of seedling roots of black mangrove inoculated by a filamentous cyanobacteria

چکیده
An isolate of the filamentous cyanobacterium Microcoleus sp. was obtained from black mangrove aerial root (pneumatophore) and inoculated onto young mangrove seedlings to evaluate N2-fixation and rootcolonization capacities of the bacterium under in vitro conditions in closed-system experiments. N2 fixation (acetylene reduction) gradually increased with time and reached its peak 5 days after inoculation. Later, it decreased sharply. The level of N2 fixation in the presence of the plant was significantly higher than the amount of nitrogen fixed by a similar quantity of cyanobacteria on a N-free growth medium. The main feature of this root colonization was the gradual production of a biofilm in which the cyanobacterial filaments were embedded. Visible biofilm production increased with time until it completely covered the entire root system of the plant. The in -and-out movement of cyanobacterial filaments from the biofilm probably allows colonization of uncolonized portions of the root several days after the initial inoculation. This is, to the best of our knowledge, the first report of the artificial inoculation of cyanobacteria on marine mangroves.

Cyanobacteria and black mangroves in Northwestern Mexico: colonization, and diurnal and seasonal nitrogen fixation on aerial roots
Nitrogen fixation and colonization by associative cyanobacteria in the aerial roots (pneumatophores) of black mangrove trees was evaluated in situ at Balandra lagoon, Baja California Sur, Mexico, for 18 consecutive months. Year-round vertical zonation of cyanobacterial colonization was determined along the pneumatophores. The bottom part close to the sediment was colonized mainly by nonheterocy...

Two isolates of the diazotrophic filamentous cyanobacterium Microcoleus chthonoplastes, one obtained from black mangrove (Avicennia germinans) pneumatophores and one from cyanobacterial mats, were inoculated onto young mangrove seedlings to evaluate nitrogen transfer from the bacterium to the plants under in vitro conditions in closed system experiments. Total nitrogen and N incorporation in pl...
متن کاملBacterial N2-fixation in mangrove ecosystems: insights from a diazotroph–mangrove interaction
Mangrove forests are highly productive ecosystems but represent low nutrient environments. Nitrogen availability is one of the main factors limiting mangrove growth. Diazotrophs have been identified as key organisms that provide nitrogen to these environments. N2-fixation by such organisms was found to be higher in the mangrove roots than in surrounding rhizosphere. Moreover, previous studies s...
متن کاملDetermination of the best concentration of cyanobacteria to improve soil fertility in rangeland (Case study: Jamabroud catchment)
Cyanobacteria play an important role in increasing soil fertility as one of the biological soil crust in rangelands. They have a significant effect on improving soil fertility because of nitrogen and carbon fixation. In this study, the effect of different concentrations of cyanobacteria as one of the soil biomasses and finally to determine the best concentration for optimizing soil fertility in...

Discoveries in recent years have disclosed the importance of marine cyanobacteria in the context of primary production and global nitrogen cycling. It is hypothesized here that microbial mats in tropical coastal habitats harbour a rich diversity of previously uncharacterized cyanobacteria and that benthic marine nitrogen fixation in coastal zones is substantial. A polyphasic approach was used t...
